Following the standard protocol, gene amplification is performed by PCR, so plasmid preparation or vector constructions are not required.
 
If primers can be designed with using leading 18mer and tail 18 mer of  ORF, gene amplification by PCR can be done and PCR product is used for transcription and translation reactions.

Therefore, vector construction and cloning are not necessary.
