No products or can not detect at all

Please check primers and reagents including detection
(e.g. whether EtBr with high staining is used )

 

1st PCR product can be slightly detected

  • increase extension time
    While  recommended extension time for 1st PCR is 30 sec / kb, but if  amplification efficiency is poor, try 1 min / kb for extension time.  For DNA up to 1 kbp,  1 min extension time could work.
  • increase number of cycles to 35

 

2nd PCR product can be slightly detected

  • set the same extension time of 2 and 3 seg. as 1st PCR program.
  • set the number of cycles of 3 seg to 35 ( up to 40)
