All-in-One kit comes with high fidelity, high efficency, high speeed DNA polymerase enabling 2 step PCR cycle, in addition to transcription and translation reagent.

The convenience of transcription and traslation remains the same, and PCR can be performed and generate template more accurately and quickly.

For 2 step cycle PCR, you do not need to worry about cycle conditions if the primers are 20 mer or more and the Tm value is 63 ° C * or pver. The PCR time can also be significantly reduced. This kit contains enough PCR enzymes for 60 reactions in 50 ul of PCR. (* Tm value must be calculated by nearest neighbor method)

eg.) 2 step PCR program for 1st gene specific primers

Predenature: 94℃, 2min
(following 30 cycles)
   Denature: 98℃, 10 sec
   Extension: 68℃, 1 min + 30 sec/kb *
(* if target is less than 1 kbp,  1 min.  if target is more than 1 kbp, 1 min + 30 sec/kb duration)

PCR reagents are designed to be used and annealed at 68° C and to be able to eliminating many problems such as non-specific amplification. So you do not need to investigate.
